Generate Biomedicines

Generate:Biomedicines (Generate Biomedicines Inc.) is an American biotechnology company headquartered in Somerville, Massachusetts, founded in 2018 by Flagship Pioneering. It applies machine learning to design and optimize proteins for therapeutic use, with a focus on immunology, oncology and infectious disease. The company describes its approach as "Generative Biology" and its technology stack as The Generate Platform, which it says can create medicines on demand across multiple therapeutic modalities rather than discovering them through conventional trial-and-error screening.

The platform is organized as a continuous generate–build–measure–learn loop: algorithms produce protein sequences targeted at a specific therapeutic question, those sequences are synthesized as physical proteins at scale, measurement systems characterize their molecular properties and functions, and the resulting data retrain the models. Reported capabilities include de novo generation of protein–protein interactions (including antibodies against defined epitopes and agonist antibodies to cell-surface receptors), optimization of enzyme properties such as processivity and specificity, and recoding of therapeutic proteins to reduce or eliminate immune recognition. Modalities span antibodies, peptides, enzymes and antibody-drug conjugates.

The pipeline has included GB-0669, a monoclonal antibody against a conserved region of the SARS-CoV-2 spike protein that entered a first-in-human dose-escalation trial in July 2023; GB-0895, an anti-TSLP antibody for asthma engineered for extended half-life and reduced dosing frequency, described by the company as in Phase 3 evaluation (SOLAIRIA study); and GB-4362, described in an August 2026 company blog post as advancing into clinical development. Reported pipeline breadth has been cited as 17 programs (2023) and more than 15 candidates.
